GMP-based fermentation method
Fermentation, gum removal, adsorption of potassium dihydrogen phosphate 12.5kg, magnesium chloride 1.2kg, glucose 2.5kg, yeast 170kg. Keep the yeast at 28-30 ℃, adjust pH = 7 with sodium hydroxide, ferment for 4h, the yeast starts to precipitate, filter to remove the yeast, and obtain enzyme solution. GMP is dissolved in water into an 8%-10% solution, 9 times the enzyme solution is added, fermented at 28-30 ℃ and pH 7 for about 3 hours, filtered at the end of the reaction, 2% diatomite is added to the filtrate, stirred and filtered, the filtrate is diluted with water to a GTP concentration of 0.3%, and adsorbed by strong alkaline quaternary ammonium I anion exchange resin column (717).

Elution and re-adsorption resin adsorbates are eluted with 0.04-0.07mol/L sodium chloride solution and 0.04-0.07mol/L hydrochloric acid, and the eluate is re-adsorbed with 717 resin.

Elution, decolorization, precipitation and drying of the secondary resin adsorbent are first eluted with 0.04-0.07mol/L of sodium chloride and 0.01mol/L of hydrochloric acid, then eluted with 0.5-1mol/L of sodium chloride solution, the eluate is added with a small amount of activated carbon and diatomite, stirred for 40min, filtered, the filtrate is added with 3 times of ethanol, let stand, filtered, the filter cake is washed with anhydrous ethanol and anhydrous ether, and the finished product of sodium guanosine triphosphate is obtained by vacuum drying.
